Does TI have reference design for two or multiple TPA3221? I will modify it by our product requirement.

Hello,
As far as I know, there is no reference design for this application. One AMP device can be set to master mode and the other can be set to SLAVE1 or SLAVE2 mode to benefit system noise performance. Please find more detailed info in the section 9.3.2 in the datasheet.
