You will require both. The EMU Boost is required for updating firmware and adding any files you wish to add to the external serial flash, for example HTML pages or certificates.

You would need to port the drivers to this platform. There are details to explain how to do this.

I have no idea what you have in mind, but if you want to make life easy, then I'd recommend the CC3200 LaunchPad - http://www.ti.com/ww/en/launchpad/launchpads-connected-cc3200-launchxl.html#tabs

The Booster pack dimensions are 50.8 X 43.2 mm.

You can also refer to assembly file available under the SDK (cc3100_boost_assy_fab_rev3p3).
