The differential input refers to one input relative to the other and not with respect to above or below supply ground. In other words if AINP is +5V and AINN is at ground, the result would appear as full-scale positive. If the AINP input is ground and the AINN is +5V then you would see a negative full-scale result. So the result will be based on the AINP input relative to the AINN input. In the first case AINP is above AINN and in the second case it is below. This example assumes that the supply and reference are 5V.
