1. No special power sequencing required.
2. A clean power supply can guarantee the achieve the performance specified in the ADC DC, thus a LDO is preferred and better than a noisy DC-DC converter.
3. Whatever your input is 0-5V or 4-20mA (converted to a voltage), the AIN_xGND should be connected to the GND directly or by a resistor for a balanced-input filter. The voltage between AIN_xGND and GND should be kept in the range -0.3V to 0.3V.

The voltage between AIN_xGND and GND should be kept in the range -0.3V to 0.3V, hence the AIN_xGND pin should be connected to the GND.

No, the ADS8688 can only support single-ended input signal, notice that this ADC can support bipolar single-ended signal (+/-10.24V,+/-5.12V,+/2.56V), also supports unidirectional single-ended 0-10.24V and 0-5.12V, so please check if your signal is a true differential or single-ended signal.
If your signal is true differential signal, plese refer to the solutions recommended in the threads on the E2E forum below:
