Hi team,
My customer want to know if this circuit could work?
When CPU read the PCA9539, one is connect to the CPU, the other is connect to the I2C Bus Splitter.
Will there be any conflict?

Move the other PCA9536 to the I2CA bus, so that one of them can be disconnected at all times.
Is there a reason why the other sensor is behind the switch?

Clemens is right! You can move the PCA9536 that is directly connected to the CPU to the I2CA bus. This will allow switching capabilities between the two devices with the same I2C address. It won't affect the other sensor because different addresses (0x40, 0x41). If you can't do that, maybe consider muxing between the two PCA9536's (Single Pole - Double Throw 2-Channel switch should do).
