The EEPROM on my board now is programmed before. The mannual attached to the program said can refer to the TUSB8040 datasheet for instructions on
externally entering programming mode, but I can't find any description related. Could you help to find it out? Thanks.

Hi,

If the hub detects an un-programmed EEPROM (value other than 55h) at byte 00h, the hub will enter Programming Mode and a Programming Endpoint within the hub will be enabled. This is for the TUSB8041.

What if the EEPROM is already programmed as 55h at byte 00h? Is there a way to enter the Programming Mode without replacing it with a new one?

Unfortunately no there is no other way to enter programming mode as this is checked right after power up.
