If I am interpreting your question correctly, you are asking whether the protection diode is connected in series or in parallel. Please correct me if I am wrong, but I have provided a few images that should be helpful.
The image below shows the ESD diode should be connected in parallel with the protected IC/device.
I am not sure if you were specifically asking about ESD751 but hopefully the above application diagram helps explain where this device fits into the system.

I am afraid your interpretation is not I meant. Anyways, the diagram you have attached helps to put the question more clearly. The circuit shows the ESD diodes are connected across the line and signal return.
My doubt is, do the ESD diodes are connected between line and earth(i.e chassis).

My apologies for interpreting your question wrong. Yes, for ESD diodes it is best to connect the ground pin to the ground chassis. This shunts current away from the sensitive device in your application. Please refer to the following E2E posts for more clarification: link1, link2
