    If you are looking at U3 is the reference design, you will not be able to replace it with the ISO7421 without making many more changes to the reference design.

    U3 is feeding back an analog voltage across the isolation barrier. This signal is proportional to the output voltage at TP3, and is seen by the CTL pin on U2 as an analog voltage. The ISO7421 is a digital isolator and therefore would not be able to send proportional feedback and only, high or low signals.
