Schematics look okay overall. For U42, note that with DIR high, the operation would be for A to B similar to the below:
For all units, note that pull-ups are not recommended with devices with the bus-hold feature (see 9.3.4 of the datasheet) and would suggest the non bus-hold alternative SN74AVC4T245. See An Overview of Bus-Hold further clarifying how the bus-hold works.
If the pull-ups are less than 30Kohms, the AXC family (SN74AXC4T245) is a newer alternative, thanks.

Hi Michael,
The U42's Operation is based on the Both cases i.e., to work on Both A to B and B to A. The 1DIR is pulled high to VCC for the Operation condition of A to B, and 2DIR is pulled low for B to A.
