Due to unavailability of TPS54627, we are trying to see if TPS54628 would be a drop-in replacement.

I think the difference is that TPS54628 supports the Eco-Mode, which allows higher efficiency during lighter load conditions.
Otherwise, they are identical. Would appreciate any inputs on the comparison.

Yes, they are drop-in alternatives. The only difference is the light load mode. TPS54628 supports Eco-mode while TPS54627 with FCCM-mode. Eco-mode has higher efficiency but larger Vo ripple at light load, while FCCM-mode has smaller Vo ripple but lower efficiency at light load.
