I noticed the datasheet mentioned the maximum regulation current is around 22mA, rather 30mA. Is it because the test condition is 20mA full scale?

Yes, your understanding is correct. This spec is used to describe the current accuracy, so its range is related to the programmed full-scale current.

Can you help clarify is there any function difference/impact if we set the Auto-Headroom enable vs Auto-Headroom disable? I checked the different value of 0x02 and 0x12, the LEDK voltage will be different.

Hi Jaden,
The headroom voltage is to ensure the current sink could be capable of expect current.
So if you disable the auto-headroom, the IC will force the headroom voltage to a larger value to make sure the current could be regulated accurately.
If you enable this function, then IC will determine the headroom voltage according to the LED current, thus will reduce the power loss on current sink itself.
