Let me confirm the possibility of the TPS628501-Q1 outputting a negative voltage.

Synchronous DC/DC converters can output negative voltages in some cases.

Does the TPS628501-Q1 generate negative voltage?

For example, when a charge remains in the output capacitor,

there is a case where a negative voltage is generated when the power supply is started again and turned on from the switch on the Low side.

I am not entirely sure if I get your question correctly.

Do you mean if the voltage will be negative if EN is pulled low? If so, No.

Also, please have a look at section 8.4.6 - Vout Discharge feature.

We understand that this power supply has a discharge function.

I'm talking about when EN switches from ON to OFF and from OFF to ON in a short period of time.

Change the way I ask questions.

From which switch is this power supply IC turned on?

In the case of low-side, we believe that the back electromotive force is generated by the low-side FET and inductor, which may generate a negative voltage.
