Are you asking if we can support creating a DRP reference schematic for this device?
I don't believe this is something we can support, however, we do already have EVMs and example schematics in section 8.2 of the device datasheet that can be used to help determine how to set this device up. I would recommend looking at those:

The DFPEVM is closest in terms of setup, however, there are still some differences that are important keep mind of, such as VBUS_DET.

I can look at the schematic on my side. Any adjusted schematics that can be sent through PM would be nice as well. In the meantime:
Have you adjusted the EQ settings on the TUSB501 at all? It could be that adjusting these settings will help the signal.
Can you send us the layout of the board being used via PM?

Should I connect to the Tx or Rx on the HOST side?

I believe it should be TX to TX in this case. From what I can tell, it looks like the system is outputting signal to the TX pins of a USB-C connector.
If possible, could you help illustrate with a block diagram the system? I am having a hard time looking through the schematic to figure out where exactly the signal is coming from, and where it is going. If you could provide something like that, or explain the systems application, it would help me determine whether it should be TX to TX, or TX to RX.

It looks like the TUSB501's are flipped. Otherwise, I believe this looks correct.
