Hi,

For the DisplayPort pathing.

1) The aux bus does not need 1M and 100k PUP and PDOWN, but it is required to have an AC coupling capacitor.

remove R25, R28. replace R525, R526 with 100nF cap.

remove R60, R63. ReplaceR527, R528 with 100nF caps.

The HPD and main link routing looks ok. Make sure that no other AC coupling is present on the main link connections. Another cap on the main links will add a series capacitance with the 100nF and push the total line capacitance to outside of the DP spec. 

Please follow up with an updated schematic.

My colleague will response shortly with feedback regarding the USB pathing.

Thank you for your suggestion, I follow up the AC coupling,  and keep the option for PUP and PDOWN, we won't install them except we need it for future, attached the updated schematic.

Thank you for your suggestion,  I have follow up your comments in the files, but for the ESD, because the display port is a internal port in our product, it will change into HDMI port for output, so we haven't add any ESD diode for it, but for all the output ports, we have already add that.

Please have the 1M pulldown on AUXN and 1M pullup to 3.3V on AUXP populated. 

It looks like you are controlling TUSB1064 in GPIO mode, but I don't see the PD controller controlling the CTL0 and CTL1 of both TUSB1064, is this correct?

Yes, we would like to set TUSB1064 in GPIO mode, and USB + DP are must in our product, so we just set CTL0 and CTL1 to enable USB+DP by pullup.  is it ok?

No, you would need to use PD controller GPIO pins to control both CTL0 and CTL1.

Yeah sorry for the confusion. I meant to say you should not have BOTH 100k and 1M connected.
As David mentioned you will need the 1M resistors PUP and PDOWNs.
