I want to ues TPS54821 to generate 5VDC at VOUT, VIN=12V, due to the need for testing, I need to supply a external 5V directly to VOUT, VIN will be powered off, so can I do this? Would it damage the TPS54821? Thanks.

Yes, you can. Please read the paper as below. You may need to add an input diode to block the reverse current flowing backwards through the IC from output to input.
