That diagram doesn't really tell me much. Can you post your input voltage, output voltage and current requirements?

Input voltage will range from (12V - 48V ) 12V nominal power supply.

Vout (5V to 24V)

Current depends on the loads connected to the ECU (but max upto 6A) , the KL31 (neg) pin of the ECU is what I am not sure whether it needs a power supply also. (to send the negative voltages fb, back to the Supply ckt)

I hope I made it partially clear , as I need to also fix whether I need both pos and neg voltages at the Loads...

Using Inverting Buck-Boost regulator topology will work ?

You are not going to get 6 A from TPS54361-Q1. You state that your output voltage is 5 V to 24 V. is it variable? What negative voltage do you need?
