Please find the attachment for RDS(on) test.

1. Whether forcing Low voltage (assume 100mV)in FB pin, will turn on Main switch, and force voltage and measure current in the SW pin will result the rds on?
2. Whether forcing high voltage (assume 600mV) in FB pin, will turn on rectifying switch and force voltage in SW and measure current in VOUT pin will result the rds on?
Please clarify ??

TI measures the Rdson by special test mode.
for customer, you can can calculate the Rdson by measuring the current and voltage of the MOSFETs when they turns on for each cycle.

How to control the each cycle? As i asked previously, is it through the FB pin? Please share your idea on the RDSon measurements in user mode.

refer to the TLV61220 EVM user guide, let the device operating normally, the device will switch. set the load current to 50ma. then measure the voltage and current.

While nominal operating mode, device's both FET (Main and rectifying switch) will be toggling, whether will it give correct measurement of the RDS?

That is a good way for customer to test RDSON.
The correct measurement is depend on your scope and probe’s accuracy.
