For the above evaluation board, understood the board is able to step up voltage to 30~85V

1. it it possible to create step up voltage +/-30 v~ +/- 85v? What I need to do to achieve it?

2. Can the +/-30 v~ +/- 85v be adjustable for example to +/-70v?

After doing that, I can adjust the voltage output by apply voltage to JP2-Vo_ADJ on the TPS61391EVM-058?

Hi Pang:

Yes, it's similar with EVM. But the regulation of the negative output may not as good as the positive port. (It's also shown in the application note)
