When the multi-cell voltage exceeds 5.6V, what is the voltage of pin REGIN? How to ensure the PIN REGIN do not exceed the max volt 5.5V ?whether iT meas the picture as follows indicates the situation that multi-cell was not exceed 5.6V?

Hi Iong,
You should refer to the schematic in the EVM User Guide here: https://www.ti.com/lit/ug/sluu904a/sluu904a.pdf There are jumper setting to use a source follower circuit in the case where the stack voltage is higher.
Best regards,
Matt
