How can I mandatory activate the balance on specific cell during board testing???

It is not possible to turn on cell balancing for a single cell. This is handled by an algorithm that balances all cells and will only do it once an OCV is taken to calculate the delta capacity.

If I power up the board then cell voltages are not the same. For example 3 cells at 3.3V and forth cell at 4V. Will it causes bq40z50-R1 to activate balance on forth cell?

The gauge will need to take an OCV measurement to calculate the delta capacity in order to trigger cell balancing.

The gauge has to be in RELAX mode in order for an OCV measurement to take place. This is controlled by the Impedance Track algorithm. Please refer to section 7.2 in the BQ40z50-R1 TRM.
