So far, I can control the TS3A227E EVM board and the basic function is work.
But I still have two questions about the TIP pin and switch. Could you help to clarify it?
2. What are the switch-OFF resistance or Isolation performance for these switch (RING2GNDFET, SLEEVEGNDFET, RING2DFET, SLEEVEDFET)?

1) Yes, you could you CPU_GPO to control the I2C register and trigger the accessory detection algorithm of the TS3A227E. You may leave the DET_TRIGGER pin floating if you will manually control the triggering. However, you must connect the TIP pin to the tip of the audio accessory for the TS3A227E to properly conduct its detection sequence.
Here is an excerpt from Detecting Selfie Sticks Using TI Aduio Jack Switches TS3A227E and TS3A225E which explains more detail why the tip pin must be connected.
2) The isolation of the depletion FETs in the TS3A227E are excellent for audio signals. What type of isolation do you need for your system?

Yes, you may use a GPIO pull down the DET_TRIGGER pin to ground creating a high to low signal that will initiate the TS3A227E detection sequence.
