If the Windows Device Manager is showing the emulator just fine, then it is possible that:
- target configuration may be messed up;
- the device itself is not able to connect due to a hardware failure or dip switch setting.
For the first scenario you will need to carefully inspect the .ccxml file and see if you selected the exact device and the emulator (Stellaris In-Circuit Debug Interface connection).
For the second scenario:
If it is a hardware failure you can test the same install with a different board to confirm the failure. If the second board works, you will have to analyze and probe the circuit for voltages (Vcc, Vcc) the crystal frequency, etc. That can be an easy or very involving job, but unfortunately it is the most critical issue that can happen to a board.
Specifically for the Launchpad, make sure you have configured the dip switches and straps correctly. Make sure that the dip switch is in the "Debug" position and the Vdd jumper is in place.

Try the LM Flash Programmer Debug Port Unlock Utility. It is at the "Other Utilities" tab of the LM Flash Programmer. After successful unlock, load a know working example program. See, if you are still getting the same error.
Possible that the last program you loaded has caused this error. Details why are mentioned at my signature link below "JTAG Communication Failures".

When you press "unlock", the process will begin. There will be instructions to hold reset button of your launchpad.

Good, that you already recovered your launchpad, and be able to use it again.
Now, you have to know why it was locked. Possible that the previous program you load to it before caused it to be locked. Some causes are mentioned at the "JTAG Communication Failures" post.
