Speaker spec 2W/4 Ohm, Expected AMP Gain: set to 20dB Please help tell me how to design AMP? And how to calculate the demand value of Speaker and what should be the input of AMP?

The schematic looks fine. Sine the speaker spec power is only 2W and PVDD is 12V, so you should limit audio input to avoid output exceeds speaker spec. PLIMIT only limits Vpeak of output, it can not limit output power.

I do not know the audio input source level, customer should handle input level circuit.

PLIMIT is fine.
