Hi Roy,
The full scale input of the ADC is 1.414Vrms so for single ended, there is a 6dB boost to get .707Vrms up to 1.414Vrms.

When an analog input is configured for single-ended use, yes, there is a 6dB boost. As mentioned above, this is get achieve a full scale input to the ADC.

I still have questions. In my measurement, we input the single ended 10mVrms. The output signal may be 6bB(internal)+32dB(set), 10m*2*40~800mVrms
But my result is 400mVrms. Do i have any misunderstanding?

Apologies for the delay here. I have been out of office the last few days and have just returned.
Doing the math, for SE I would expect ~-5dB and for differential ~-11dB with 32dB of gain applied. Your calculations look correct as with mine, no errors were accounted for.
