1. the ADC offset calibration is commonly done at start up of the device. It helps to calibrate the device to a known reference point and leads to more accurate measurements. it is also commonly done when temperature changes have occurred, this helps to eliminate any drift. you can think of this as an internal offset calibration, meaning for the device. it is natural for ICs to drift with time or temperature, performing an offset calibration reduces this for the ADC.
2. Yes, this method would help to calibrate the system. I would suggest performing the sensor calibration after the ADC offset calibration. you can think of this calibration as an alingment between your input and the ADC.
