the AFE2256EVM FPGA board only works with both frontend boards COFa and COFb connected, is this correct?
Is there a way to work only with one COFa or COFb adapter board mounted?

yes, your observation is correct. It is due to the FPGA firmware design, not related to the device itself.
The FPGA waits for valid data from both devices. Hence, EVM does not capture data with single device. To avoid this issue, you can modify the FPGA code to ignore 2nd device data; or simply remap both data FIFOs to single device output).
