Which windows version are you running?

This GUI has been verified and tested on Win 10 machine. I feel there is some issue with the serial port selection in the GUI

Can you go to Options → Serial Port, and change the port to the other available port with the (Texas Instruments) or ACCtrl tag. One of the two ports with these tags will connect to the hardware

Did you follow the correct user guide procedure? Why did you load blinky.bin?
Also what do you mean 2 USB ports are connected to EVM?

Please follow the procedure outlined in the EVM user's guide and revert back.

Why did you load blinky.bin?

-> Because I want to test the MSP-EXP432E401Y Board that it can work properly.

No need to connect the 2 USB. Can you tell me where in user guide, it is mentioned , you need to connect 2 USB cables?

We need to remove the already connected USB to connect  the other side for firmware update only.

It needs to connect 2 USB cables in this picture.

And, I have tested that if only use the U7 Port, it can not show the 2 XDS110 Port(COM6 & COM7) in PC Device Manager list.
