When you are using the USB OTG can you even connect with the EVM GUI such as below? Did you check your device manager if the USB device pops up?
Can you try using a laptop with just a normal USB cable?
UART is normally for debug purposes and not used as the main form of communication.

I already use the EVM GUI that communicate successfully. But I want to control using our external MCU like arduino directly. Could I use this way using another port on 471TP EVM?

DLPC6540 controller in the DLP471TPEVM inly supports two interfaces for commands i.e USB and IIC.
