Thanks for using the E2E forum! We should be able to get you a clear answer to your question.
"Trigger Out 2 Supply" is essentially the reference voltage for "Trigger Out 2" output trigger. There are jumper pins on the EVM as seen in the image below taken from the LightCrafter 4500 User's Guide Figure 1-11 that can be used to modify the reference voltage, and therefore the trigger voltage. These jumpers (J13 and J15) allow you to chose the output trigger voltage level.
The LightCrafter 4500 User's Guide has more information on this topic in section 7.2 "Output Trigger Connectors".
