Are you using DLP4500 EVM? You want to replace FYEEYE between LED and DMD by a diffuser?
The FLYEEY is an integral part of illumination optics and its purpose is to image LED source uniformly on the DMD.
Could you please explain in detail why you want to use diffuser? Also drawing of optics?

Yes correct.

Diffuser could also spread out the light, and if it could replace flyeye, then it will be cost saving.

Can you share how the diffuser would fit in this case to the system? A lens array (fly's eye) is the typical method of homogenization as shown in our optical engine reference design example:
