TCA9509: TCA9509 connections

Tool/software:

for our design we have couple of PCBAs with different voltage rails.  I am planning to use TCA9509 to support I2C bus across both boards .  I have following I2C tree with 2 options.  I would like to know which option would you recommend.  Also are there any pull ups required on SIDE A of these devices?
