When CMC is placed on the top layer, isolation is maintained in top layer (4-layer board). Whether same isolation needs to be followed in the rest of the laye's too.

Thanks for reaching out. On a CAN bus, a CMC is used to remove common mode noise. A CMC does not provide isolation. The ISO1050 maintains isolation from side 1 to side 2 of the PCB. When a CMC is used with the ISO1050, it is only connected to the CAN bus side of the device, and does not interfere with isolation in any way.
