About the isolation audio circuit shows as the right image in Figure 2-1, I want to know if the grounds at both ends need to be connected together externally to form a loop after being isolated?

For example, they are connected through the outer metal shell? If a loop is not formed, will the audio not be output?

    Thank You for reaching out. Glad to see this ISOUSB application note coming to use for your application of interest. Now coming to your question:

    NO, the 2 isolated grounds need not be connected in any way for the successful operation of USB Peripheral Audio Output. Please refer Figure 3-2 from the same document below:

    Moreover, If you connect the 2 grounds in any way, like outer metal shell, then it will nullify the advantage of using the ISOUSB in this application as it creates a Ground Loop path again.
