Can DRV2667 drive 6.8uF piezo speaker for 20-20kHz (audio output) @50Vpp?

No, for 6.8uF at 50 Vpp you will be limited to something below 1kHz.

Perhaps the IC LM48580 would be more suited, this device is intended more as a Piezo speaker driver.

LM48580 is rated for 30Vpp. Would DRV5825P be suitable for this application?

Yes, that would be suitable, the DRV5825P also has the benefit of an integrated algorithm capable of piezo speaker protection and acoustic tuning
