my question is would this affect I2C communication? since their I2C report is NACK now

TXP/RXP will not affect I2C communication, maybe SCL/SDA pin failed too. Pls contact local CQE for quality sopport.

do you see any potential risk that may damage TX1n and TX1p on the layout ( red circle is TX1n,p)
they may face some EIPD symptom, any recommendation for this?
layout looks normal ,are there ESD protection device on TX1/RX1?, if not , plug in/plug out could damage the device.

I understand TX/RX may suffer EIPD, but SDA/SCL is internal signal I can't see why it's also malfunction.
and you said these two are not related.
any idea why I2C NACK? or any signal you still want me to check other than TX/RX?
If the ESD damage in Vdd , the damage could affect other pins too.
