I want to generate +2.5V with respect to the negative voltage of the reference.
For example, add 2.5V to -4V to generate -1.5V, -3.5V to generate -1V.

Any noise on -4V (connected to the GND pin of REF50) will directly appear on the output, and it will not be a clean supply which is why this scheme is not preffered.
You will get stable output with respect to gnd in the app note method suggested by Masoud.
