I am designing a ioniser, can I use a PSR flyback IC with a high turn ratio transformer to create a -10kV output?
If this does not make sense, please give me some advises about this application.

I don't think that we have tested such a high voltage application with this device; or any device.
The transformer may have large parasitics that could interfere with the control of the regulator.
You may need to go with some kind of voltage multiplier.

Do you mean add a voltage multiplier after output of FSR flyback?
Could you provide some reference design to describe more detail about it?

I am not sure that we can provide details about your 10kV.
It may be best to search for other on-line resources that can
provide a starting point for your design. Then TI can help with
selecting any regulators you may need.
