Hi Xia,
yes, all capacitance connected to the output must be taken into account. The recommended typical output capacitor value of 2 × 10-μF or 1 × 22-μF should be placed close to the device pin as seen on the layout example in the datasheet.

Hi Xia,
unfortunately, it is not possible to predict the impact of the bead from the schematics. I recommend that you measure the total capacitance at the Vout pin, if the effective total capacitance (without bead) is larger than 47μF.
