No, a shorted interwinding turn on the primary will at best result in a decrease in the secondary winding volatge.

Output voltage could increase or decrease depending where the short occurs. Maybe check, how one sample compares to another and if a difference is observed then investigate how a shorted turn might have occurred (high voltage breakdown, moisture, wire varnish breakdown, etc).
