I'm using the TPSM63606 and noticed the TPSM63606S variant. Is it literally the same part except for the SS functionality (ie. pin compatible and all else is the same)? How do you configure the SS functionality and is it on by default?

Spread spectrum is something that's trimmed in/out of the part at the OPN level. You need to order the part with Spread spectrum enabled to get the feature.

I guess what I'm trying to ask is, can I technically use the TPSM63606SRDHR as a drop in replacement for the TPSM63606RDHR if the former is available but the latter is not? This is to mitigate parts shortage issues.
