Is it necessary to make an battery reverse polarity  protection with the TPS6101x ?

    it depends on your application requirement. the TPS6101x would be demaged if the input polarity reverse, because a negative voltage is applied to the device input pin. maybe you can place a diode in series with the battery to prevent negative voltage.
