In general you do not want to apply a voltage on the EN pin when there is no voltage on the VIN pin.
There is the possibility that you will turn on a parasitic ESD diode on that pin.
If this is a requirement, you will need to limit the current with a resistor.
I would try to limit the current in the range of 100uA or so.

Yes, you can set the EN voltage high prior to applying the input voltage.
