Can TPS2549-Q1 support reverse current protection? If the input is short to ground while the output is clamping at 5V due to the output capacitor.

If the device is enable , the TPS2549-Q1 don't support reverse current protection.
If the device is disable , the device can block the current from output (the FET internal body diode will not impact as it has been connect to ground).

In both scenario, will the reverse voltage (Vout>Vin) or current damage the device?

Hi Dongbao,
The device internal haven't the reverse current limit function , If apply the voltage in Vout the current should come from OUT to IN pin. In this case this voltage will charge to front end DCDC output. The device internal has thermal protection. When big current come from OUT to IN pin it will cause the OTSD.
