Can we directly connect Blank pin to ground?
We are using only one IC and controlling it through I2c.

It is OK to connect BLANK to GND. By this way outputs would change immediately when you latch data. But I am doubt that I2C protocol can be used to communicate with IC, since the slave would not have ACK signal back to your controller.
Could you help to share more information about your application, like the number of LED per channel and the current for LEDs? Maybe we have other I2C device which can be recommended for you.

So you do not use TLC59282 as LED driver right? Could you help to share what application you designed for? If you need 16 channels to control 16 bit data, I think you could take a look at LP5018, which have 18 channels with 12-bit PWM outputs individually. You can set them to FFh/0 as on/off.
