I have questions about the layout for LM5165.
1. Do we need to have the power ground and signal ground?
2. If so, should the power ground connect to GND first or the signal ground should connect to the GND first?

I went through the layout section for LM5165. It did not mention to have the power ground and the signal ground, so if that's the case, then there is no need to have the power ground and signal ground, just to have one ground for all pins should be good. Is that correct?

Yes, you can have just one GND plane on the board but it is advised to keep the power stage on one side of the IC and keep the sensing and sensitive nodes on the other side as shown in the layout example. This effectively isolates the power stage AC current GND noise from the sensitive nodes.
