I am interfacing PRG1 RGMII Port 2 with two MACs—between the AM6442 and the AM263P4 controller—in a MAC-to-MAC configuration.

Could you please confirm whether MDC and MDIO connections are required in this setup, or can they be omitted?

Hello Dilip,

MDIO pins can be omitted in that case. It is an I2C-like interface used to change settings and see status of a PHY. I think you will need to modify software driver not to use MDIO as it will expect to see some registers and it will not find any in your case. For the latter I cannot recommend anything because I'm not familiar with software.
