Hello Annie,
You could keep a corner reflector or strong metallic object in front of the radar and look at the received power level of the object peak in the FFT with and without the radome. The difference will provide you are loss in the radome.

generally speaking: do radomes effect phase measurements as well or do they only attenuate? If not, DoA estimation is distorted, correct?

you are right , they could impact the phase as well. Depends on the shape of the radome, proximity of the sensor etc. So yes, doing amplitude and phase measurements at different angles will be a good way to evaluate.
