Can you provide the input capacitance of the SN74CBTLV3257 when the mux is enabled? All I see in the datasheet is when the MUX is not enabled. I am assuming Ci = 3pF, but want to confirm.
What's the difference between Ci and Cio(off)?

The on capacitance/ Cio on of the device should be around 16pf but this is a estimate.
In reality if on capacitance is a concern for your application would using the sn74CB3Q3257 as a p2p replacement be a option?
The quick difference between these device are
The CBTLV has <=200Mhz BW and CB3Q has 500Mhz BW .
The CBTLV is rail to rail output with 3.3Vcc , only 0V to 3.3V i/O can be passed through whereas CB3Q is beyond rail to rail output where with 3.3Vcc, 0-5V I/O can be passed through.
Ci is control input capacitance. And Cio is input/output terminal capacitance when the signal path is disabled. Here is also another good appnote that helps explain CBTLV datasheets like this one
