CC2541: cc2541: BTTool Port not select

hy Marie, 
No i did not program my device with host test.. what is its purpose?

I programmed my device with host test But i am unable to select port is SerialBootTool.
How can i select port without smartrf05 board.?
Can i do the same work using cc debugger?

As i have no serial port.

You cannot use CC Debugger. You would need to use serial-to-USB cable to connect CC2541 UART pins to desktop for BTool to communicate with if you don’t have SmartRF05.

1-Kindly tell me which configuration i have to select to connect cc2541 with Btool.

2-I uploaded CC2541_SmartRF_HostTestRelease_All.hex  and tried to connect it with BTool But i got this error

Kindly suggest solution for this.

1-I  programmed cc2541 HostTest(CC2541EM-SBL) .

2-I connected the cc2541 with serial port  like this.(provided 3.3V externally making ground common)

3-Now when i connected it with serial port and open BTool got this error(Also there is no device in list).

Where i am making mistake kindly mention.

Is this level shifter ok ?

And at HV what should I apply?(5V from where?) 

OR is this OK?

I checked the BLE pins(Tx,Rx) there is constant (3.2-3.3)volt at Tx and on Rx(0-1) volt. When I open the Btool it remains same.When I connect the probs i get this data(may be garbage).
