Hi I am using the MIFA antenna for ESp32 module. I have designed the antenna as per the ESP32 Wroom module but I can see all teh EVm boards use 1oz copper thickness. I f I want to increase the thickness to 2oz what wil be the effect on antenna. I am not using any ground layer or any plane below antenna.

There should be no measurable effect going from 1 oz to 2 oz copper thickness.

Thank you Jake for confirming. One more concern, I am using Isola 370HR for my PCB and thickness of PCB will be 1.6mm. is that fine?

Yes Isola 370HR is fine (similar to what's specified in the design, DE104). If you're changing the thickness of the PCB this can affect the antenna resonant frequency. You will want to simulate to see if any adjustment needs to be made.
